There is no sound from the speakers connected to my Windows XP-based desktop. All the wires are connected securely. How can I troubleshoot this problem?

The answer to a similar query as posted by you is http://www.askiyogi.com/hardware-peripherals-drivers/6773.html
Please follow all the steps mentioned here:

(a) At the back of your system, there may be multiple ports for output sound, if you have a sound utility software installed, it will let you configure these ports, check what configuration this port has in which you have plugged the speaker. The default one has a green color.
(b) If you have any other audio output option like HDMI, Optical out etc, check if these are selected as default output in the player that you are using.
(c) Check if the sound in muted in sound control panel for default sound out device.
(d) Try the troubleshooter option given in the link http://windows.microsoft.com/en-IN/windows/help/no-sound-in-windows
(e) Reset the BIOS to default.
(f) Install the latest service pack if missing
(g) Run the windows update
(h) Run a full anti spyware scan
